Why not copy ONLY install.wim to the NTFS partition, as all other files will be copied to the FAT32 partition?

What does "c." stand for? The drive letter?
    Circa? "a) fat32 size approximately 1GB"

    Just guessing.

For some reason, it does not always work with just install.wim. it only takes a few seconds extra to copy the lot.

    c. means approximately (it means circa - Latin). It is similar to e.g. or i.e.

The abbreviation is used a lot in UK.

    2) copy all files from iso to ntfs partition
    3) copy all files EXCEPT install.wim to fat32 partition.
Done as you said above, but I got the following message when I booted it from the USB drive, whose first and second partitions are in FAT32 and NTFS respectively.

Is this because I don't have the following?

Why are you not using Option #2 and just run Rufus? It works. Now that you've got your Thumb drive partitioned in Fat32/NTFS, it may need to be put back to standard FAT32 only. Sometimes that can be hard to do, or impossible without tricky formatters.

Some using 7-Zip to extract the ISO contents directly to the Thumb. Again, all FAT32 format.

Some mount the ISO and copy to thumb. Again, all FAT32 format.

Because Rufus will erase all files on my 28 GB USB drive.

I am using a fake drive for testing, and it is very easy to reformat it. I have done so several times.

Yup! I've used Rufus as long as I've been making my own ISOs. Follow the instructions and it just works.

I bought four 8 GB USB drives for a few bucks and have dedicated those to my ISOs.

I've never considered using a bootable flash drive as multi-purpose.
